DPPC Selectively deuterated at the choline alpha and beta segments as described by Gally et al. (1975). Nondeuterated DPPC was obtained from Fluka, Switzerland

Sample protocol Lipid (~30 mg) was dispersed in ~30 mL of buffer at 50 °C and centrifuged to form a pellet. The pellet was transferred to a NMR sample tube and measured in the presence of excess buffer. A second series of experiments used a constant lipid concentration of 70 mM. No differences in quadrupole splittings were detected between the two sample types.

DETAILS The interaction of mono-, di-, and trivalent metal ions (Na+, Ca2+, La3+) with DPPC bilayers was investigated. Quadrupole splittings were evaluated from powder-type spectra. The alpha segment was found to be almost insensitive to temperature, while the beta segment showed a pronounced decrease in splitting with increasing temperature The study states that only relative signs could be determined. However, based on established databank conventions for PC headgroups, alpha is positive and beta is negative.
