Reagent sources
POPC Choline headgroup is alpha- and beta-deuterated. POPC synthesized from corresponding PA (Avanti Polar Lipids, Birmingham, AL, USA) following the method of Harbison and Griffin (1984). Alpha- and beta-deuterated choline synthesized as described in Gally et al. (1975) and Seelig and Gally (1976).

Sample protocol Mixtures of headgroup deuterated POPC and POPE were prepared by pipetting corresponding volumes of concentrated stock solutions (10 mg/ml in dichloromethane/methanol) into NMR measuring tubes. The solvent was evaporated and lipids were dried under high vacuum (under P2O5) for several hours. Phospholipids were dispersed at room temperature in 20 mM Hepes (pH 7.4) buffer containing 150 mM NaCl, prepared with deuterium-depleted water. Lipid/buffer ratio was 1:2 (%w/%v).

Recycle delay of at least 7 T1. Alpha and beta order parameters interpolated from Fig. 5A and 5B respectively at Xb=0.099 using WebPlotDigitizer. SCD = 0.00784 * delta_vQ(kHz).
